吉846/掖3189 RIL群体遗传连锁图谱加密和玉米丝黑穗病抗性相关QTL分析
本研究基于实验室前期构建的吉846(高抗)/掖3189(感病)含273个家系的F7重组自交系(RIL)群体的连锁图谱,进一步筛选多态性的SSR标记加密图谱,并结合3年抗病鉴定结果,对玉米抗丝黑穗病进行QTL定位。结果表明,将亲本间存在差异的66个新SSR标记加密到遗传图谱中,构建了含160个SSR标记和49个AFLP标记的遗传连锁图谱,覆盖玉米基因组3302.8cM,平均图距15.8cM;应用完备区间作图法共检测到3个抗丝黑穗病相关QTL分别位于染色体bin2.09、bin3.04和bin9.04区域;利用混合线性模型法检测到7个未报道的抗病相关QTL,分别位于染色体bin2.05、bin6.02、bin8.05、bin9.01、bin10.03和bin10.07区域。本研究结果为抗玉米丝黑穗病分子标记辅助育种提供理论依据。

In our previous studies, the RIL population derived from Ji846 (high resistance) and Ye3189 (susceptible) containing 273 lines was constructed and we obtained a genetic linkage map. In this study, the density of linkage map was increased by SSR markers. QTLs for head smut resistance were analyzed based on 3-year disease-resistant identification results. The sixty-six SSR markers were added to the previous genetic map of RIL population. The new map contained 160 SSR markers and 49 AFLP markers. The total length of the new map was 15.8cM, and average interval distance was 15.8 cM. Complete interval mapping (ICIM) was used in QTL analysis combined with the incidence of head smut RIL population. Combined with average incidence of head smut in three years, three QTL were detected on chromosome bin2.09, bin3.04 and bin9.04 area. Interaction of field incidence test results in three years and environment were analyzed by mixed linear model approach. Seven new QTLs were obtained located on bin2.05, bin2.08, bin6.02, bin8.05, bin10.03, and bin10.07 area. The results will be benefit for maize head smut marker-assisted breeding.
