本研究以与郑58回交多代并自交纯和的Z58Ab-t1为材料,以其背景亲本郑58(Z58)为对照,首先利用Bt-Cry1Ab/Ac金标免疫试纸条进行检测,初步证实目标Cry1Ab-t蛋白得以表达;其次进行抗虫性鉴定,表明该转化子与对照相比具有良好的抗虫性,这也进一步证实目标蛋白的表达;在此基础上,用酶联免疫(ELISA)法研究不同地区、不同组织器官和同一组织器官不同生育期Cry1Ab-t蛋白表达的动态,结果表明不同地区、不同组织器官和同一组织器官不同生育期Cry 1Ab-t蛋白表达均有显著不同。不同组织器官间,叶片表达量最高,苞叶表达量最低;同一组织器官不同生育期Cry1Ab-t蛋白表达为灌浆期>抽丝期>拔节期>苗期。

Dynamic expression of bt protein in transgenic maize was studied, using Z58Ab-t1, as the materials, and their non-transgenic genetic background Inbred line, Z58, as control. Firstly, The expressing of cry1Ab-t protein in Z58Ab-t1 was confirmed by Jin-Biao immune detection strips and insect-resistance identification. Secondly, the expression of cry1Ab-t protein had significant difference in different areas, growth stages and organs, was verified by ELISA method. It showed that the highest expression of cry1Ab-t protein was in the leaf of corn, while the husk leaf had the lowest expression. Furthermore, the expression rule of cry1Ab-t protein in different growth stages was filling stage> silking stage> jointing stage> seedling stage.
