四倍体青贮玉米染色体加倍及检测技术研究

以辽单527、辽单青贮178、辽原一号三个玉米品种为试验材料,通过6个浓度的秋水仙素溶液处理,进行染色体加倍,并采用流式细胞仪检测加倍效果。结果表明,当秋水仙素浓度达0.1%时,三个品种加倍率均达到最高值,其中辽单527的加倍效果最好,最高可达36.3%;利用流式细胞仪能够快速准确的检测染色体倍性,OTTO法的检测效果较好。 英文摘要: There varieties of silage maize including Liaodan 527, Liaodan 178 and Liaoyuan 1 were chosen as matirials and 6 concentrations of colchicine were used for chromosome doubing in this work, aimed to detecte the double effect by flow cytometry (FCM). The results showed that: the doubling rate were all reached the highest level when the concentration of colchicine was 0.1%, and Liaodan 527 worked best, the doubling rate was 36.3%; FCM could correctly detect the chromosome multiple and OTTO method had better effect.
