玉米CLC家族基因差异表达及其与 NO3-、Cl-吸收之间的关系初探

为了研究玉米硝态氮吸收与CLC基因表达量之间的关系,首先通过对该基因家族的7个基因的蛋白序列比对,发现它们具有比较高的同源性,之后在玉米的苗期用KNO3、NaCl、PEG对其处理,分别在处理1、4、8、16、24、36小时后检测其体内NO3-、Cl-的浓度,同时取上述几个时间段处理的玉米根和叶,用qPCR检测CLC家族基因的表达量。综合玉米中NO3-、Cl-的变化与CLC家族基因表达量的变化,明确了在KNO3、NaCl、PEG处理下CLC家族基因表达量变化的规律,发现BT066984.1、GQ412103.1、NM001176231.1和NM001136935.2这四个基因对硝酸盐的响应明显,很可能具有运输硝酸盐的作用。这为深入研究玉米CLC基因家族对硝态氮素的吸收指明了方向。 英文摘要: In order to study the relation between the nitrate nitrogen absorption and the amount of CLC gene expression in maize, firstly, based on the protein sequence alignment of seven genes in this protein family, we found that they have relatively high homology. Then treating by KNO3, NaCl and PEG at seedling stage, tests the concentration of NO3-, Cl- in the corn body after 1h, 4h, 8h, 16h, 24h, 36h of the treatment respectively. At the same time selecting the roots and leaves treated by the above-mentioned time periods, tests the amount of CLC family gene expression with qPCR. By synthesizing the change of NO3 -, Cl and the quantity of CLC family genes expression in maize, confirmed the change rule of the amount of gene expression in CLC family by the treatment of KNO3, NaCl and PEG. We found that, BT066984.1、GQ412103.1, NM001176231.1 and NM001136935.2, these three genes response to the nitrate obviously and are likely to have the effect of transportation for nitrate which indicates the direction for the further study of the nitrate nitrogen absorption by the CLC family gene in maize.
