玉米淀粉QTL定位分析

试验以5M017和5D07为父母本杂交组配的110株F2代分离群体为试材,筛选68对多态性较好的SSR引物构建遗传图谱,覆盖1607.7cM,平均遗传距离23.64cM。用完备复合区间作图法(ICIM)对玉米淀粉(总淀粉、直链淀粉、支链淀粉)含量进行QTL分析。结果表明:共检测到17个与玉米淀粉含量有关的QTL,其中10个与总淀粉含量有关,分布在除第4以外的染色体上,有8个位点加性效应为负值,总淀粉含量增效基因主要来自父本;与直链淀粉含量有关的QTL有5个,分别位于第2、3、6、8、10 染色体上,4个位点加性效应为正,直链淀粉增效基因多数来自母本;与支链淀粉含量有关的QTL有2个,位于第3、10染色体上,均表现遗传负效应;位于第10染色体标记区间phi059-umc1432检测到三种淀粉共同存在的QTL。 英文摘要: Abstract : A genetic linkage map was established, based on 110 F2 individuals from the cross between parent 5M017 and 5D07. The total genetic distance of the genetic linkage map was 1607.7cM with an average of 23.64cM between two makers. Quantitative trait locus controlling maize starch (total starch, amylose, amylopectin) content were analyzed by using integrated composite interval mapping (ICIM) method. The results showed that 17 QTLs related to maize starch were identified. 10 of QTLs were related to total starch content, which distributed on the chromosomes except the 4th, the additive effect of 8 sites were negative, so the synergistic genes of total starch content was mainly from male 5M017. There were 5 QTLs about amylose, which located on chromosomes 2,3,6,8,10, and the synergistic genes of amylose content were mainly from female 5D07 because he additive effect of 4 loci were positive. 2 QTLs related to amylopectin were found which located on chromosomes 3,10 and the effects value were negative. A QTL controlling all of the three maize starch content were detected on phi059-umc1432 of chromosome 10. 查看全文
