玉米杂交种产量相关位点分析及优异等位变异挖掘

【目的】本研究利用分子标记筛选产量相关位点,估计位点基因型值,挖掘产量相关的优异等位变异位点,并分析高产组合的等位变异位点构成,为筛选携带优异基因位点材料提供参考。【方法】采用NCⅡ设计,将13个玉米自交系配置成42个杂交组合,选用120对SSR引物进行分子标记,采用基于回归的单标记分析方法,通过表型变异对标记变异的回归分析,筛选玉米产量相关等位变异位点,估计等位变异的效应和位点的基因型值,剖析杂种组合等位变异的位点效应。【结果】筛选得到phi089、umc1142和umc1700等8个标记位点,及其phi089-4、umc1142-5、umc1700-3、phi102-1、phi047-1、umc2317-2、bnlg1892-5和bnlg1352-3等8个优异增效等位变异位点,bnlg1352-3/5、phi089-4/7、umc2317-2/5、umc1142-4/5和bnlg1892-1/5等5个基因型值较大的位点为等位变异的优异杂合位点。构成杂合位点的2个等位变异位点各自的加性效应及其显性效应共同影响着杂合位点的基因型值。P4、P5、P10和P12等材料为优异亲本。【结论】筛选得到多个优异位点及其载体亲本,为指导玉米自交系亲本组配提供理论与技术支撑。 英文摘要: Abstrect:【Objective】 The study attempts to screen yield-related locus by molecular markers, and estimate the locus genotype value, and select the locus-allel associated with yield, and analyze the composition of the locus-allel in great hybrids. It will provide the information for selecting the carrier materia of great locus-allel.【Method】The NCII design was used to configure 13 maize inbred lines into 42 cross combinations, and 120 pairs of SSR primers were selected for molecular markers. The method of regression analysis based the phenotypic variation on marker variation is used to select the great locus, and estimate the effect of locus and its genotype values, and analyze the effect of heterozygous locus on of the hybrid. 【Result】There are eight great sites are significantly associated with yield (phi089, umc1142, umc1700 etc),along with their eight great locus-allel(phi089-4, umc1142-5, umc1700-3, phi102-1, phi047-1, umc2317-2, bnlg1892-5, bnlg1352-3). There are five great heterozygous locus with larger genotype values (bnlg1352-3/5, phi089-4/7, umc2317-2/5, umc1142-4/5, bnlg1892-1/5). The value of genotype is both related to the dominant effect value of the locus and the additive effect of the allele pair in heterozygous locus; P4, P5, P10, and P12 is the great parent materials.【Conclusion】There are some great locus and their carriers were sifted in the analysis,it can be used to guide the mating of maize inbred lines.
