抑制ZmCol3基因表达调控玉米开花期研究

ZmCol3基因是玉米CCT基因家族的一个重要成员,候选基因关联分析研究结果表明ZmCol3基因与玉米开花期紧密连锁,然而调控ZmCol3表达是否可以有效改良玉米开花期至今仍然未知。本研究采用ZmCol3基因RNAi载体构建、农杆菌介导玉米遗传转化、转基因材料开花期表型鉴定等研究方法,评估抑制ZmCol3基因表达对玉米开花期的影响。转基因玉米基因组PCR结果证实人工合成RNAi片段已成功整合到玉米基因组中;qRT-PCR结果表明在不同转基因玉米株系中ZmCol3基因表达受到不同程度的抑制;温室转基因玉米开花期相关性状调查结果表明抑制ZmCol3表达,可以将玉米抽雄、散粉和吐丝时间提前2-3天。上述研究结果证实ZmCol3具有调控开花期的生物学功能,抑制该基因表达进而缩短玉米开花期可以作为行之有效的方法应用到玉米熟期改良研究中。 英文摘要: ZmCol3 gene is an important member of CCT gene family in maize. Association analysis for candidate gene shows that ZmCol3 gene is tightly linked with maize flowering stage. However, it is still unknown whether maize flowering stage was improved effectively by regulating ZmCol3 expression. In this study, ZmCol3 gene RNAi vector construction, agrobacterium mediated genetic transformation and phenotypic identification of transgenic maize at flowering stage were used to evaluate the effect on ZmCol3 inhibited expression at maize flowering stage. PCR showed that RNAi vector had been successfully integrated into the maize genome; qRT-PCR results showed that ZmCol3 gene expression in different transgenic lines were inhibited in different degrees; Traits in the survey at flowering stage showed that inhibition of the ZmCol3 expression, could make the corn tassel, pollen and silk 2-3 days ahead of time. These results confirm that ZmCol3 has biological function in regulating flowering stage, inhibiting the ZmCol3 expression and shortening the flowering period of maize can be used as an effective method to improve the maturity. 查看全文
