长枝木霉菌株GAAS L3-1-0.8对玉米形态学指标影响及生物学特性
为了解长枝木霉菌株GAAS L3-1-0.8对玉米的促生作用及生物学特性,为下一步发酵培养和有效利用提供技术支撑。采用该菌株不同浓度的悬浮液处理玉米种子,结果表明:与对照相比,1.0×106 CFU/mL对玉米发芽率、芽长、根长、根数和根鲜重呈显著差异(P <0.05),有明显的促生作用。该菌株GAAS L3-1-0.8的生长温度范围为10~35℃,菌丝最适宜扩展温度为25~30℃,最适产孢温度为25℃;菌落在pH值为5.0~ 0.0的培养基上能够迅速扩展,菌丝最适扩展的pH值为6.0~9.0,产孢最适酸碱度为6.0~7.0;营养生长较好的碳源为葡萄糖、麦芽糖和乳糖,最适宜菌丝扩展的碳源为葡萄糖和麦芽糖,在含有葡萄糖碳源的培养基上产孢量最高;营养生长和产孢量最适氮源为酵母膏;24 h黑暗条件下营养生长最快,12 h 光暗交替条件有利于产孢。

To understand the promoting effect and biological characteristics of Trichoderma longibrachiatum strain GAAS L3-1-0.8, in order to provide technologic support of fermentation conditions and efficient utilization, adopting different concentration of T .longibrachiatum spore suspension to treat with corn seed, The results indicated that 1.0×106 CFU/mL of this strain GAAS L3-1-0.8 could significantly promote corn seed germination rate, germinal length, root length, root numbers and root fresh weight compared with control(P <0.05). T. longibrachiatum GAAS L3-1-0.8 could grow under 10~35℃, pH 5.0~10.0, with optimal condition at 25 ~30 ℃, pH 6.0~9.0, and the largest conidia germination at 25 ℃, pH 6.0~7.0. Mannitol, Maltose and Lactose were the better carbon source for hypha growth, but dextrose was the best carbon source for spore bearing. While yeast extract was the optimum nitrogen source for mycelia and sporulation. The best cultural condition was 24h under dark, but the largest sporulation quantity for mycelial was 12 h light and 12 h dark.
