应用直接PCR技术快速筛查转基因玉米

以转CP4-epsps基因玉米为研究对象,以玉米内标准基因zSSIIb及三种常见转基因成分(CaMV35S启动子、NOS终止子、CP4-epsps基因)为检测靶标,建立了基于直接PCR技术的玉米转基因成分筛查方法。该方法不用提取纯化DNA,只需取直径约为0.5 mm的叶片加到PCR反应体系中进行扩增,与常规PCR方法相比,在保证结果准确性的同时,大大缩短了检测时间,提高了工作效率。该方法具有操作简便、结果可靠等优点,适用于玉米叶片中转基因成分的快速筛查。 英文摘要: In this study, the transgenic maize with CP4-epsps gene was selected as the research material to detect the endogenous reference gene zSSIIb and three common transgenic elements (CaMV35S promoter, NOS terminator and CP4-epsps gene), through which a screening method for maize transgenic components based on direct PCR was established. Using this method there was no need to extract and purify the DNA. We only took 0.5mm leaves in diameter and added it to the PCR reaction system for amplification. Compared to conventional PCR method, the developed direct PCR method greatly shortened the working hours, and improved work efficiency, as well as ensuring the accuracy of the results. The method has the advantages of simple operation and reliable result, and is suitable for rapid screening of transgenic components in corn leaves.
